Tretinoin is a metabolic intermediate product of vitamin A in vivo, which mainly affects the growth of bone and promotes the proliferation, differentiation, and the metabolism of epithelial cells. For the treatment of acne, psoriasis, ichthyosis, lichen planus, pityriasis rubra pilaris, hair follicle keratosis, squamous cell carcinoma and melanoma tumor disease.

Dosage and usage

Oral, 10mg/ times, 2 to 3 times / day. For external use, 0.05% to 0.1% cream or ointment. Local inunction, 1 ~ 2 times / day.

Pharmacology and Toxicology

Pharmacology: epidermal keratinocytes, melanocytes and dermal fibroblasts are important targets for the role of vitamin A acid. Retinoic acid can influence melanoma cells, melanin formation, its role is a number of points, on the activity of tyrosine hydroxylase (th), dopa oxidase and 2 hydroxy indole oxidase type three enzyme catalysis have inhibitory effect, so as to reduce the formation of melanin, reduce skin pigmentation. The vitamin A acid had no effect on the activity of tyrosinase and melanin in normal human melanocytes. When the skin is aging, or by drugs, UV radiation and trauma damage, vitamin A acid can correct or prevent harmful factors to the dermal connective tissue biochemical composition and morphological structure caused by abnormal, to stimulate skin cell matrix protein synthesis, accelerate the formation of new connective tissue in the dermis, and can improve the tensile strength of the wound. The effect of vitamin A on collagen synthesis in normal skin was not affected. In addition, the inhibition of the A acid on the cell chemotaxis, which plays an anti-inflammatory role. All trans retinoic acid A has no direct effect on the sebaceous gland and its secretion. Toxicology: oral retinoic acid on experimental animals, including mice, rats, hamsters, rabbits, monkeys and other and people have a strong teratogenic effects. Skin topical retinoic acid on the sensitive period of embryonic development in the mouse, rat, hamster, rabbit mother have clear embryo toxicity and teratogenicity, and can lead to maternal toxicity. Retrospective data so far found no cause of human skin after topical teratogenic. D A acid on the skin irritation.
